The Lotz Lab is studying mechanisms and treatments of intervertebral disc degeneration (IDD) which affects a large population of older individuals and represents the major cause of back pain. The research on disease mechanisms uses human spines and genetic mouse models to investigate changes in cells, genes and proteins that develop during the aging process and the development of IDD. These studies examine cell function, RNA expression patterns using RNA sequencing and single cell RNA sequencing. In a newly funded program about mechanisms of joint tissue destruction and joint pain, we are using Omics technologies (RNA-seq, single cell-RNA-seq, ChIP-seq, epigenetic analyses, spatial transcriptomics) to identify dysregulated transcription factors and test candidate genes in vitro and using mutant mouse strains. A staff scientist position is available in the Paust lab at TSRI to perform NIH-funded research on NK cell receptor diversity, immunological memory, and NK cell responses to HIV. Recent discoveries by the Paust laboratory and others indicate that subsets of human NK cells mediate adaptive immunity to HIV-encoded antigens; however, NK cell receptor repertoires diversify throughout life, and high NK cell receptor repertoire diversity is associated with increased risk of HIV acquisition. These data present a challenge for vaccine design, as both protective NK cell-mediated memory responses and potentially harmful NK cell receptor repertoire diversifications are consequences of antigen exposures.
